Insulin - like Growth Factor I Expression by Tumors of Neuroectodermal Origin with the t ( 1 I ; 22 ) Chromosomal Translocation

چکیده
Expression of insulin-like growth factor I (IGF-I) mRNA by some tumor cell lines of neuroectodermal origin has been described. To further explore the significance of IGF-I mRNA expression in these tumors, a more extensive analysis was performed. Most (9 of 10) neuroectodermal tumor cell lines with a t(11;22) translocation (primitive neuroectodermal tumor IPNETI, Ewing's sarcoma, esthesioneuroblastoma) expressed IGF-I mRNA, whereas 0 of 15 cell lines without the translocation (PNET, neuroblastoma) expressed IGF-I. Furthermore, inasmuch as all neuroblastoma (12 of 12) cell lines examined expressed IGF-II RNA, the pattern of IGF expression could distinguish between these closely related tumors. CHP-100, a PNET cell line with the t(11;22) translocation, was shown to secrete both IGF-I protein and an IGF binding protein, IGFBP-2. This cell line also expressed the type I IGF receptor mRNA, and blockade of this receptor by a monoclonal antibody (aIR3) inhibited serum-free growth. These data demonstrate that IGF-I expression is a property of neuroectodermal tumors with a t(l 1;22) translocation and that interruption of an IGF-I autocrine loop inhibits the growth of these tumor cells. ANTISENSE RNA TO THE TYPE I INSULIN-LIKE GROWTH FACTOR RECEPTOR REVERSED THE TRANSFORMED PHENOTYPE OF PC-3 HUMAN PROSTATE CANCER CELL LINE IN VITRO
The insulin-like growth factor I receptor (IGF-IR) plays an essential role in the establishment and maintenance of transformed phenotype. Interference with the IGF-IR pathway by antisense causes reversal of the transformed phenotype in many rodent and human tumor cell lines. We stably transfected the PC-3 human prostate cancer cell line with an IGF-IR antisense RNA expression plasmid. The ...
متن کاملP-64: The Relationship between Polymorphism in Gene of Insulin-Like Growth Factor-I and The Serum Periparturient Concentration in Holstein Dairy Cows
Background: One of the most important metabolic factors affecting the reproductive activity is insulin-like growth factor-I (IGFI) concentration changes after calving. Recently, Maj et al. (2008) discovered a significant association between the IGF-I genotypes based on the 5'-untranslatedregion (5'-UTR) of IGF-I gene and the IGF-I blood level. The objective of this study is to investi...
متن کاملPrenatal Diagnosis of Triploidy in Fetus with Unexpected Chromosomal Translocation of Maternal Origin
Triploidy is a lethal chromosomal abnormality. Foetuses with triploid condition have a tendency to die in early conception and very few survive to term. In this study, we report the prenatal diagnosis of fetal triploidy with unexpected chromosomal translocation. A 27 years old women was referred to our clinical cytogenetic department due to history of previous conceptus with intrauterine growth...
